“Evaluation Of Several Phenotypic Methods Of Antibiotic Susceptibility Pattern For The Detection Of MRSA With Molecular Profiling RT-PCR With The Detection Of MecA Gene”

INTRODUCTION:- Methicillin-resistant Staphylococcus aureus (MRSA) is a well-recognized public health problem throughout the world and community MRSA strains are now epidemic in India others countries. The resistance rates of S. infection multidrug resistant increasing, making clinical anti-infective treatment diagnosis more difficult.
 AIM:- To evaluate several phenotypic methods antibiotic susceptibility pattern for detection with Molecular Profiling Rt-PCR MecA gene.
 MATERIALS AND METHODS:- present study was prospective observational carried out at Department Microbiology, Santosh Medical College, Ghaziabad tertiary care hospital over 12-month period from August 1, 2020 to July 31, 2021. Both IPD OPD sample comprised 385 isolates as various specimens were included. Antibiotic by Kirby-Baure disc diffusion performed according Clinical Laboratory Standard Institute (CLSI) guidelines. Detection like, Cefoxitin Disk Method, Oxacillin method, E-Test method gene RT-PCR out.
 RESULT:- In 384 samples, majority found be (E-Test) strip 114 (29.7%), followed cefoxitin 113 (29.4%) oxacillin 99 (25.8%). However, we observed high incidence other antibiotics such Erythromycin 265 (69.0%), Clotrimozole 228 (59.4%), Tetracyclin 144 (37.5), Vancomycin 102 (26.6%) Refampicin (26.6%). We also that Linezolid 354 (92.2%) Teicoplanin 325 (84.6%), Gentamycin 272 (70.8%), Clindamycin 249 (64.8%) Amoxyclave 281 (73.2%). this molecular test mecA pus 36 samples blood 17, urine 33, Sputum 05, pleural fluid 04, wound swab 16, Vaginal 06, CSF 01, Throat 2.
 CONCLUSION:- gold standard assay determining methicillin PCR technique detecting gene. But Because still time intensive expensive, it not yet available 95% routine laboratories choice where E-test reliable than drug can utilised on regular basis better results.
